What is/are Nilotinib?

Nilotinib (AMN107, trade name Tasigna), in the form of the hydrochloride monohydrate salt, is a tyrosine kinase inhibitor approved for the treatment of chronic myelogenous leukemia.

Medical uses

Nilotinib was approved as Tasigna in the USA and the EU for drug-resistant chronic myelogenous leukemia (CML). In June 2006, a Phase I clinical trial found nilotinib has a relatively favorable safety profile and shows activity in cases of CML resistant to treatment with imatinib (trade name: Gleevec [USA]/ Glivec [Europe/Australia/Latin America]), another tyrosine kinase inhibitor currently used as a first-line treatment. In that study 92% of patients (already resistant or unresponsive to imatinib) achieved a normal white blood cell counts after five months of treatment. The drug carries a black box warning for possible heart complications. The use of low doses of nilotinib is being investigated for use for Parkinson's and Alzheimer's disease, as well as for ALS, dementia and Huntington's disease

Adverse effects

  •     Headache with chest pain and severe dizziness, fainting, fast or pounding heartbeats;
  •     Fever, chills, body aches, flu symptoms, sores in your mouth and throat;
  •     Pale skin, weakness, easy bruising or bleeding;
  •     Blood in your urine or stools;
  •     Severe pain in your upper stomach spreading to your back;
  •     Nausea, loss of appetite, dark urine, jaundice (yellowing of the skin or eyes);
  •     Lower back pain, numbness or tingly feeling around your mouth;
  •     Urinating less than usual or not at all;
  •     Muscle weakness, tightness, or contraction;
  •     Fast or slow heart rate, weak pulse, feeling short of breath; or
  •     Sudden severe headache, confusion, problems with vision, feeling like you might pass out.

Other common side effects may include:

  •     Diarrhea, constipation;
  •     Mild skin rash, temporary hair loss;
  •     Headache, back pain, joint or muscle pain;
  •     Tired feeling; or
  •     Cold symptoms such as stuffy nose, sneezing, cough, sore throat.
